Who Takes Custody of Remaining Retatrutide Material When a Nonclinical Study Pauses or Ends?

When a nonclinical retatrutide study pauses, ask the contract research organization (CRO) to confirm what it holds, which portion is a designated reserve and who may authorize the next move.

At closeout, record the instruction and the actual receipt or disposition separately so the team can see what remains in custody.

Two researchers reconcile sealed blue-capped vials with a paper inventory and handoff record.

Establish what the CRO still holds

In a hypothetical handoff, a project manager sends a pause notice assuming the external facility will keep all remaining retatrutide stock together. The custodian puts unallocated working stock on the routine clearance list because the study file only flagged the archive sample for retention. Ask the custodian to reconcile the physical count with the original record and identify which containers still need an authorized instruction.

Retatrutide remains investigational. The US FDA states that it is not a component of any FDA-approved drug. The handoff discussed here concerns material already held by an institution for an authorized nonclinical study.

Reserve samples, working stock and study records

Surplus working material is the unused portion of a lot after experimental work stops. A designated reserve or analytical sample is a portion of the batch retained for later evaluation. Record the reserve balance separately. Archived test-item documentation and inventory logs serve a different purpose from physical samples and follow their applicable retention schedule after surplus is cleared.

US 21 CFR Part 58 covers specified nonclinical studies supporting, or intended to support, FDA research or marketing permits. For covered studies lasting more than four weeks, §58.105(d) sets a good laboratory practice (GLP) requirement. Reserve samples must be retained batch by batch for both test and control articles, as provided in §58.195. Four weeks refers to study duration. Under OECD GLP guidance, an analytical sample is retained from each batch except in short-term studies. Its archive period takes account of stability, whether evaluation is still possible, and national requirements.

A pause needs a custodian and a review date

A study pause with unresolved custody needs an identified custodian and a review date. When testing stops pending sponsor review, working material may lack a final disposition directive. The custodian can log a pending hold under the existing study arrangement, with the open question and the person who will follow up.

At final study closeout, designated reserves may still need to remain in custody. For the working surplus, check the study plan, agreement and facility procedures for continued custody, an agreed return or transfer, or documented disposal. For a proposed transfer, obtain receiving-party acceptance before routing the material. Record the actual receipt when the handoff takes place.

Who can authorize the next step?

Check the study agreement for authority to issue disposition instructions. Clear closeout records specify three controls.

  • Authorization. Name the sponsor project manager, study director, or quality representative empowered for this study to issue disposition instructions.
  • Pending action. Record the interim custodian, the unresolved custody question, and the review trigger for the next review.
  • Evidence of execution. Identify the record reconciling the ledger. Use the completed disposal record under applicable facility procedures or the actual dated receipt from the receiving party. Reconcile the quantity and unit before closing the entry.

Single-lot handoff slip

To initiate handoff, the project manager can email the CRO custodian with the single-lot handoff slip. State whether the study is paused or has reached final closeout. Ask the custodian to identify the designated batch reserve, the remaining working surplus, the current custodian and any instruction awaiting sign-off. Request execution evidence for the study file. Separating instruction and authorization from physical execution and receipt keeps an open request visible until the inventory is reconciled.

Study and physical inventoryEnter authorized study ID, investigational retatrutide lot code, and CRO inventory ledger reference. Add the physical quantity and unit by container and record any discrepancy.
Material categoryDesignate surplus working material or batch reserve sample, noting study records are archived separately. List reserve and working portions separately if they share a lot.
Study status and current custodianMark study pause with unresolved custody (interim hold) or final study closeout. Identify the current custodian and location.
Open custody questionIf paused, state the open custody question, its owner and the next review date or trigger. Include a local hold code if used.
Disposition instructionChoose continued custody, agreed return or transfer, or disposal under facility procedures. Record an interim instruction if the final choice is still open.
Instruction and authorization sign-offRecord the authorizing signatory, role, authority under the agreement or study plan, approval date and instruction reference.
Receiving party acceptanceFor authorized transfer or return, attach documented receiving institution acceptance before routing, or mark N/A.
Execution and actual receiptRecord the CRO custodian, actual quantity and unit, execution date, and archive ID for completed disposal or transfer receipt. Name the follow-up owner for any item still pending.
